Stereological Study on the Juxtaglomerular Granular Cell in Adrenalectomized Mice

Abstract
The juxtaglomerular granular cells of the mice were investigated by electron and light microscopy. For the electron microscopic study, the renal tissue was fixed by perfusion method with 2.3% glutaraldehyde in 45mM cacodylate buffer following prerinsing with heparinated 0.1% Ringer-lidocaine soltion. After the removal of the tissue, it was post-fixed in 0.1% osmium tetroxide in 45nM cacodylate buffer by immersion. For the histological study the tissue was fixed in Zenker-formol solution, and stained by Wilson¡¯s method. The juxtaglomerular granular index(JGI) and granular cell index (GCI) were estimated and the volume fraction of secretory granules and endoplasmic reticulum of the juxtaglomerular granular cell were calculated by stereomorphometry in normal and adrenalectomized mice. The results observed were as follows : 1. The JGI and GCI of adrenalectomized group are higher than normal. The increase of JGI is seemed to be due to the appearance of Type 3 cells and not by the proliferation of juxtaglomerular granular cells and epitheloid change of other cells. 2. Both of the volume fractions of secretory granules and endoplasmic reticulum shows significant increase in the experimental group. 3. In the quantitative analysis of the juxtaglomerular granular cells the stereoloigcal morphometry is seemed to be more accurate and recommendable than juxtaglomerular granular index and granular cell index.
